Two forms of PTEN with distinguishable functions on the postnatal development of eel (Anguilla japonica) gas glands
Abstract
PTEN (Phosphatase and Tensin homolog on chromosome 10), a phosphatase is associated with tumor suppression, is an important factor in embryonic development and body size control. Two forms of PTEN (PTEN-long form and PTEN-short one) have been showed in the Japanese eel (Anguilla japonica). This paper was to elucidate the possible differences on these two forms of PTEN on the development of gas glands (rete mirabile), the differences were confirmed by investigating both the expression profiles in the hormones-stimulated female eels and a primary culture of gas glands cells treated with various factors. RT-PCR and immunohistochemical staining were employed to analysis the changes of different PTEN productions. The data showed that their cellular distributions, as well as expression levels of both genes and cellular proteins content were different by various factors in vitro; the differences of their expression profiles or cellular protein contents can be distinguished in the stimulated gas glands with different tissue masses in vivo. Our data suggested PTEN-long form has a more direct role as a negative regulator of the PI3K/PIP3/PKB pathway, PTEN-long form may be a new member of PTEN homologue; the function of PTEN-short form, needed more evidences to support the idea, might involve into cellular stability.
